Koro-sensei reminisces about his first meeting with Karasuma, where they first butted heads and fought against each other, ultimately coming to respect one another. However, Karasuma eventually points out their actual meeting wasn't nearly as eventful.

Affiliation: Ministry of Defense; Japanese Government; Class 3-E Occupation: Government Worker, Assistant Teacher and PE instructor Base of Operations: Kunugigaoka Junior High School  He is a government worker associated with Ministry of Defense. He first appeared announcing Korosensei's deal with the government and their mission to assassinate him before the end of the year. He later became the assistant teacher and P.E. instructor of class 3-E to train the students.   He is well known amongst militaries for his outstanding reputation and is a powerful individual, which he has proven when he easily overpowered Irina's master, who is a retired professional hitman, and even Korosensei himself is afraid upon seeing his capability for the first time.  He is strict, but kind man, though he keeps a distance between himself and the students due to his status as an agent and his own professionalism. He is one of a few who is very perceptive enough to see Nagisa's talent as an assassin. He seems to know Korosensei's past, though he has no intention to indulge into it further.

Tadaomi Karasuma

Koro-sensei is a teacher for class 3-E of Kunugigaoka Junior High School and the main antihero of Ansatsu Kyoushitsu. He was responsible for carving out a large chunk of the moon to become a permanent crescent moon and plans to destroy the earth after teaching class 3-E for a year.  His name comes from Korosenai (æ®ºã�›ã�ªã�„) means "cannot be killed" and sensei, means teacher. Literally, his name means unkillable teacher.  Koro-sensei is made up of anti-matter. He will inevitably die in March, whether the students assassinate him or not.  It is later revealed that the chance of this happening was less than 1%.  Koro-sensei was previously known as the "God of Death."  Koro-sensei did not actually destroy the moon, a rat infused with anti-matter did.

Nagisa is seen as one of the weakest students in the class due to his small stature and mediocre physical abilities that are comparable to some of the girls in the class. However, he is actually the most talented to become an assassin out of all the students, something that acknowledged by Korosensei and Karasuma. He is able to draw closer while hiding his bloodlust to make a surprise attack and able to make his opponents falter through his bloodlust alone.  Nagisa usually writes Korosensei's weaknesses that at first seem minor (since it doesn't look like major weaknesses), but eventually revealed to be useful since once they're able to almost kill Korosensei because of one of his weaknesses that is the same as the one that Nagisa has written down in his note.  Outwardly friendly and polite to his friends and everyone, he hides a somewhat darker side of him as an assassin that won't be able to be found out during daily classes and will only shown during training and assassination time. According to Karasuma, whenever Nagisa unleashes his killing intent, he isn't training and truly goes for a kill.  In the end he is the one who bears the burden of killing Koro-sensei. 

Nagisa Shiota

She hasn't shown any capabilities during her training to become an assassin, and so far has only been shown to discuss Korosensei's weaknesses along with Nagisa. She experiences a slight inferiority complex when it comes to sex appeal, resulting her to get angry whenever there's a woman with big breasts and readily protest.   It is later revealed that she was put in class E after she broke one of the headmaster's belongings.  In chapters 128 and 129 of the manga, and episode 14 of the season 2 of the anime, Kayano reveals that her real name is actually Akari Yukimura, sister of the previous class E teacher Aguri Yukimura. She believes that Korosensei murdered her sister and stole a pair of tentacles in order to avenge her. Kayano hid her tentacles for the entire duration she posed as a student in class 3E, as well as masked her enormous bloodlust by staying near Nagisa Shiota.

When a mysterious creature chops the moon down to a permanent crescent, the students of class 3-E of Kunugigaoka Middle School find themselves confronted with an enormous task: assassinate the creature responsible for the disaster before Earth suffers a similar fate. However, the monster, dubbed Koro-sensei (the indestructible teacher), is able to fly at speeds of up to Mach 20, which he demonstrates freely, leaving any attempt to subdue him in his extraterrestrial dust. Furthermore, the misfits of 3-E soon find that the strange, tentacled beast is more than just indomitable—he is the best teacher they have ever had!

Adapted from the humorous hit manga by Yuusei Matsui, Ansatsu Kyoushitsu tells the story of these junior high pupils as they polish their assassination skills and grow in order to stand strong against the oppressive school system, their own life problems, and one day, Koro-sensei.
